OPN3 was discovered in deep brain regions, specifically the arcuate nucleus and the paraventricular nucleus of the hypothalamus, however there have been limited research in the functionality of OPN3. Oancea lab has discovered that OPN3 functions in a light independent manner and signals through Gai to negatively regulate the melanogenic G\u03b1s-coupled MC1R in human epidermal melanocytes. Neuronal melanocortin receptors (MC3R and MC4R) are found in the arcuate nucleus and paraventricular nucleus of the hypothalamus and play non-redundant roles in mediating energy balance. Both MC3R and MC4R share similar structural identities to MC1R and have been found to couple to G\u03b1s. Additionally, Cone and his colleagues found that MC4R couples to Kir7.1 in hypothalamic neurons independent of G-protein activity. Previous data in our lab has revealed that OPN3 colocalizes and forms a physical complex with MC4R. The goal of this study is to test the hypothesis that OPN3 functions to negatively modulate MC4R-mediated signaling. More specifically, we overexpress OPN3 and MC4R in HEK293 cells together with Kir7.1 and measured the potassium current at diferent potentials. We only observed a significant amount of current in cells coexpressing OPN3 and this effect seems to be occluded by MC4R addition. Nuestro blanco de acci\u00f3n son los canales de calcio voltaje operados CaV3.2. Estos canales son clave para la excitabilidad neuronal y disfunciones de los mismos han sido descriptas para patolog\u00edas graves como la epilepsia. Nuestro laboratorio ha demostrado una sensibilidad de estos canales de calcio a la actividad constitutiva de D5R.En c\u00e9lulas en cultivo de la l\u00ednea HEK293T, la coexpresi\u00f3n de D5R con CaV3.2 produce una disminuci\u00f3n de la corriente observada mediante la t\u00e9cnica de patch clamp en c\u00e9lula entera. La incubaci\u00f3n de las c\u00e9lulas con 1uM de HVA durante 44 horas, a raz\u00f3n de repetir el est\u00edmulo cada 6 horas, produce una restituci\u00f3n significativa de la corriente en presencia del receptor D5R, (valores de las corrientes, p valor, test estad\u00edstico utilizado y n\u00famero de c\u00e9lulas, adjuntados en la presentaci\u00f3n) Nuestra hip\u00f3tesis es que el HVA ocluye el efecto de la actividad constitutiva de D5R sobre la corriente CaV3.2 actuando como ligando tipo agonista inverso del receptor. Este efecto del HVA es an\u00e1logo al de un compuesto categorizado dentro de los antipsic\u00f3ticos cl\u00e1sicos con probado efecto como agonista inverso de D5R, la clorpromazina.
